The route from Berlin to Lutherstadt Wittenberg is a part of the Anhalter Bahn from Berlin to Halle. This route (until Wittenberg) has a lengh of around 95-100 kilometres, which is very long. And with building this route, there is still the gab between Wittenberg and Bitterfeld to drive from Berlin to Leipzig. For that reason, I suggest building just to Jüterbog first. This route would have a lengh of around 65 kilometres if you build until Berlin Hbf(tief). I suggest building the route until Berlin Gesundbrunnen and/or Berlin-Spandau. You may build also the S-Bahn north-south-tunnel, a part of the seperate S-Bahn-route between Gesundbrunnen and Südkreuz. After that, 2 S-Bahn-tracks follow the two main-line-tracks and goes at last around 2 kilometres seperate to Teltow Stadt (- S25/S26). Then, the route goes with 2 tracks all the time via Ludwigsfelde and Luckenwalde to Jüterbog. In Jüterbog, there are tracks in four directions, but the IC and ICE trains don't stop there. Now, look at the map:

For freight trains, we definetly need the short route from the "Berliner Außenring", just a few kilometres from Birkengrund/Großbeeren to Diedersdorf, where you could start freight trains.

If you build in the north to Gesundbrunnen or Spandau, also ICE trains will stop there. The RE4 drives to Berlin-Spandau (then to Rathenow), the RE3 and RE5 are driving to Berlin Gesundbrunnen (then to Schwedt(Oder) or Stralsund Hbf (RE3) and Rostock Hbf or Stralsund Hbf (RE5)). Some ICE trains start in Berlin Gesundbrunnen.

I didn't choose this route lengh for fun. If you build next the route Jüterbog - Wittenberg - Bitterfeld with a lengh about 65 kilometres as extention for Berlin - Jüterbog and Rapid Transit, you could drive from Berlin to Leipzig. But you may build directly the route Berlin - Wittenberg, but this has a lengh about 92 kilometres from Berlin Südkreuz and something about 100 km from Berlin Gesundbrunnen.
This route is also available for the TS2021 with Berlin-Wittenberg or Berlin-Leipzig, there is also the station Berlin Gesundbrunnen built.

My train suggestion for this DLC:
BR 112.1 with maybe modernized Dostos (4. Generation) (to explain the 'maybe' look below)
Some freight with maybe BR 185.0, BR 189, BR 193, BR 151 or some older stuff.

My future-DLC suggestion for this DLC:
BR 481 for the S-Bahn Berlin
BR 445 for the RE4 (if licensing with the ODEG is not a problem)
    Any other freight loco

My service suggestion for this DLC:
    Option 1: 2021
RE3 Berlin Hbf (maybe Gesundbrunnen) - Jüterbog every 60 minutes with 4 OR 5 Dostos
RE4 Berlin Hbf (maybe Spandau) - Ludwigsfelde every 60 minutes and to Jüterbog in rush hour with BR 445 (from DLC)
RE5 Berlin Hbf (maybe Gesundbrunnen) - Berlin-Lichterfelde Ost (maybe to Diedersdorf, but in real their don't stop any person trains) every 60 Minutes with the BR 112.1 with 4 OR 5 Dostos
ICE trains drivable between Berlin Hbf and Südkreuz (maybe Gesundbrunnen, until Jüterbog drives AI) something like every 120 minutes (some Sprinter to Frankfurt and all Sprinter to munich) with BR 403.
Freight trains from Diedersdorf to Jüterbog and a few trains from Großbeeren.
    S25 and S26 with the BR 481

    Option 2: 2012
RE3 Berlin Hbf (maybe Gesundbrunnen) - Berlin-Lichterfelde Ost (maybe Diedersdorf) with a BR 112 and 4 unmodernized Dostos (the train drove to Elsterwerda like today the RE5) every 60 Minutes
RE4 Berlin Hbf (maybe Spandau) - Ludwigsfelde/Jüterbog with a BR 112 and 4 unmodernized Dostos every 60 Minutes (the trains drove to Wittenberge and Wismar like today the RE2)
RE5 Berlin Hbf (maybe Gesundbrunnen) - Jüterbog with BR 112 and 4 unmodernized Dostos every 60 Minutes (trains are: RE5 Rostock - Wittenberg, RE5 Stralsund - Falkenberg)
Some IC trains with BR 101 and IC1 as KI to Jüterbog (and ICE with BR 401, BR 402 and BR 411)
    Freight like above
    S25 and S26 with the BR 481

    Now some pictures:

Probably that´s not necessary, as you got right besides the Anhalter Bahn the container terminal of the GVZ Großbeeren.

Excellent idea and if you want to link this with Nahverkehr Dresden at Röderau build the Jüterbog Röderau Line as a Branch that way Berlin is linked to Dresden via Jüterbog Röderau line. Basically you did link Berlin with Rapid Transit at Bitterfeld. Long term plan is Berlin Dresden Leipzig intercity network bundle this route Rapid Transit Leipzig Riesa Nahverkehr Dresden with the Elsterwerda extension and Berlin Elsterwerda.
