They're all very different, so it depends what type of services you like most.

High speed, long distance? Fast, commuter services? Freight? :)

From that list, I'd go for either ECW (East Coastway) or SEHS (Southeastern High Speed).
I did enjoy playing RRO though.

IoW is a very short route.

Of those you don’t own, I only have ECW and SEHS and I like both. Both use similar trains in the 375 and 377, but SEHS also has the 395 for HS passenger service while ECW has the venerable 66 for freight. Some of the routes you already own will allow some additional layers for freight and/or excursion tours on both routes.

If I had to bring it down to one choice, I think ECW is the much better optimized of the two as of right now as I think the SEHS needs some touch up on the high speed portion.

For variety, RRO. You have the S Bahn commuter services, and freight services.

As a bonus, it will also add S Bahn services to Koln - Aachen, so it adds more variety to another route at the same time.

I'd say since you want variety go for Southeastern high speed(since you own Tess Valley and Transpennine you got extra services and stock there) But if you want the best looking passenger route go East Coastway, It looks really good, feels good too.
And if you prefer german stuff, Rhein Rhur Osten. again, Main Spessart and Ruhr Sieg Nord add extra services there, however the route feels too short, It would be much better if they extended to Dusseldorf Hbf, add ICE services, more freight, yards and variety.

I have all of them it doesnt make any difference to me. It depends what you want to do. I like both Freight, Passenger and Shunting operations. Mosts Routes have at least one of them so they all tick my boxes.
