After initially being frustrated by discovering that brake releases only exist on one side of a freight car in the Clinchfield route, I further found that it's possible to step over the couplers: just approach them slowly until the "Press E to step over couplers" prompt appears and then (duh!) press E. This feature may also exist on other routes, but I hadn't encountered it before.
