My favourite routes for Train Simulator are the Brennerbahn, Canadian Mountain Passes and the East Coast Mainline London - Peterborough. I like all type of locomotives but my favourite for Train Simulator are the ÖBB 1293, 4023, 1142, the LNER Peppercorn Class A2 and the DB BR103 Trans European Express.
